OurMouseTREM-1ELISAKitisasandwichELISAintendedforthequantitativedeterminationofmouseTREM-1incellculturesupernates,serumandplasma(heparin,EDTA),celllysatesandtissuehomogenates.Trem1,Triggeringreceptorexpressedonmyeloidcells-1,isencodedbyTrem1gene.TheexpressionofTrem1isinmonocytesandneutrophilsbutnotinlymphocytes,dendriticcells,orothercelltypes.Trem1isa30-kDglycoproteinthatisreducedto26kDbydeglycosylation,inagreementwiththepredictedmolecularmass.TheTrem1genewhichcontains4exonsmapstochromosome6p21.1,withinaTREMgeneclusterandthemouseTrem1genemapstochromosome17inaregionthatshowshomologyofsyntenytohumanchromosome6.TheexpressionofTrem1isupregulatedbystimulationwithlipopolysaccharide(LPS),gram-negativebacteria,andfungi.Cross-linkingofTrem1onneutrophilsinducesinterleukin-8(IL8)andmyeloperoxidasesecretion,whilecross-linkingonmonocytesinducesnotonlysecretionofIL8butalsoofmonocytechemotacticprotein-1(MCP1,orSCYA2)andtumornecrosisfactor(TNF);MCP1andTNFsecretioncouldbefurtherupregulatedbyLPS-mediatedpriming.Trem1engagementalsoinducesupregulationofadhesionmolecules(e.g.,ITGB1)andcostimulatorymolecules(e.g.,CD40).Trem1isassociatedwithDAP12(TYROBP),amoleculefrequentlyassociatedwithactivatingreceptors.
